Protocol for Antibiotic Sensitivities and Analysis
Using an Equine Express II® cooled semen & embryo transport system 1. Prepare each dose of semen to the Exodus Protocol of 750 million to 1 Billion progressively motile sperm to extend to a 5:1 ratio (25 million/ml) 2. If you are testing two different antibiotics, draw two syringes of the plain extended semen as your control group. Draw up to the 50 ml line on each syringe and mark as control. 3. Prepare one complete dose of extender with Amikacin & K-Penn and prepare one complete dose of extender with Ticarcillin. 4. Draw up two syringes, one from each antibiotic to the exact protocol as stated above in step #1 5. Mark each syringe according to the individual antibiotic in each syringe. 6. Using a new Equine Express II® cooled semen & embryo transport system, place a control syringe and a test syringe in the bottom of each container. Place the Thermoregulator and a fresh and frozen special coolant pac on top, push the styrofoam lid in place and finish with placing the corrugated lid into the sides of the box securing the package according to the instructions. 7. Mark the outside of each box according to the particular antibiotic under test. 8. At 12, 24 & 36 hours, quickly pull out each syringe and place one drop of semen on a clean & warmed microscope slide from the incubator.  Analyze for progressive motility. 9. On a separate sheet of paper, write the total progressive motility of each syringe and place out of site in a folder until the entire test is finished. This will prevent a bias opinion if you know the results from the previous total progressive motility analysis. 10. With all of your testing complete, pull out all of your previous results and make your qualified decision accordingly.  Some stallions are better at 24 hours than they are at 12 and so on and so fourth.  This is why it is so important to not allow the previous test results to interfere with your total progressive motility values.
